چکیده
Nitrogen (N) availability is one of the most important factors regulating plant metabolism and growth as it affects global gene expression profiles. Dynamic changes in chromatin structure, including histone modifications nucleosome assembly/disassembly, have been extensively shown to regulate under various environmental stresses plants. However, involvement related nutrient responses has demonstrated only a few studies date. In this study, we investigated function chaperone NUCLEOSOME ASSEMBLY PROTEIN1 (NAP1) proteins N deficient conditions Arabidopsis. nap1;1 nap1;2 nap1;3 triple mutant (m123-1), N-responsive marker genes lateral roots were decreased conditions. addition, m123-1 plants showed delay deficiency-induced leaf senescence. Taken together, these results suggest that NAP1s affect

Histone Chaperone-Mediated Nucleosome Assembly Process
A huge amount of information is stored in genomic DNA and this stored information resides inside the nucleus with the aid of chromosomal condensation factors. It has been reported that the repeat nucleosome core particle (NCP) consists of 147-bp of DNA and two copies of H2A, H2B, H3 and H4. Regulation of chromosomal structure is important to many processes inside the cell. In vivo, a group of h...

In eukaryotic cells, genomic DNA is primarily packaged into nucleosomes through sequential ordered binding of the core and linker histone proteins. The acidic proteins termed histone chaperones are known to bind to core histones to neutralize their positive charges, thereby facilitating their proper deposition onto DNA to assemble the core of nucleosomes. For linker histones, however, little ha...
متن کاملOn the mechanism of nucleosome assembly by histone chaperone NAP1.
The process of mononucleosome assembly mediated by histone chaperone NAP1 was investigated using DNA fragments 146 and 207 bp in length containing the Lytechinus variegatus 5 S rDNA nucleosome positioning sequence. A quantitative description was derived using gel electrophoresis and fluorescent anisotropy data. First, NAP1-bound H3.H4 was released forming a DNA-histone tetramer complex with a t...
متن کاملNucleosome assembly protein 1 exchanges histone H2A-H2B dimers and assists nucleosome sliding.
Eukaryotic chromatin is highly dynamic and turns over rapidly even in the absence of DNA replication. Here we show that the acidic histone chaperone nucleosome assembly protein 1 (NAP-1) from yeast reversibly removes and replaces histone protein dimer H2A-H2B or histone variant dimers from assembled nucleosomes, resulting in active histone exchange. Transient removal of H2A-H2B dimers facilitat...
متن کاملThe histone chaperone Nap1 promotes nucleosome assembly by eliminating nonnucleosomal histone DNA interactions.
The organization of the eukaryotic genome into nucleosomes dramatically affects the regulation of gene expression. The delicate balance between transcription and DNA compaction relies heavily on nucleosome dynamics. Surprisingly, little is known about the free energy required to assemble these large macromolecular complexes and maintain them under physiological conditions. Here, we describe the...
